PDZ domain polarity complexes

چکیده
a member of the group, said they used 58 embryos to generate three stem-cell populations, but two were lost at an early stage. But he said this was fewer embryos than was used by other groups and attributed the success to the use of donated embryos that had been screened for genetic disorders, rather than discarded embryos from in vitro fertilisation procedures which may not be of sufficient quality. The first human embryonic stem cell lines were created in 1998. There are just nine lines available across the world, the majority from Singapore, and they are extremely expensive. Scientists at King's College were granted one of the first two licences to produce human embryonic stem cells by the Human Fertilisation and Embryology Authority early last year. " Embryonic stem cells are the most useful ones because they are very primitive — they have the potential to develop into any type of cell in the body. We have to work out how to coax them to develop into specific types of cells, " says Minger. " We know that cell replacement therapy is effective in treating diseases such as diabetes and Parkinson's but it has been very difficult to produce enough cells to treat even one patient. An embryonic stem cell line would enable us to produce unlimited amounts. " Holy grail: Stem cells are the target of much research effort and offer promise in the research and treatment of many diseases and conditions. What are they? Three protein complexes, each containing at least one PDZ protein–protein interaction domain, are critical regulators of cell polarity in animals. Each complex can be referred to by one of its primary components: the Par-3 or Baz complex, including Par-6 and aPKC; the Crb complex, including Sdt; and the Scrib complex, including Dlg. The Lgl protein, which has activities similar to the Scrib complex, seems to physically associate not with the Scrib but with the Baz complex. Are they conserved across species? Yes: orthologous complexes are found in worms, flies and vertebrates. Not only is the composition of the complexes conserved, but there is also remarkable conservation of their localization, particularly in epithelial cells. Nevertheless, the mutant phenotypes caused by defects in components of the complexes vary across species, suggesting that the story may be more complicated. How were they identified? Primarily through genetic screens in model organisms. Mutations of Crb, Sdt and Baz cause …

A molecular-properties-based approach to understanding PDZ domain proteins and PDZ ligands.
PDZ domain-containing proteins and their interaction partners are mutated in numerous human diseases and function in complexes regulating epithelial polarity, ion channels, cochlear hair cell development, vesicular sorting, and neuronal synaptic communication. Among several properties of a collection of documented PDZ domain-ligand interactions, we discovered embedded in a large-scale expressio...
متن کاملA Specificity Map for the PDZ Domain Family
PDZ domains are protein-protein interaction modules that recognize specific C-terminal sequences to assemble protein complexes in multicellular organisms. By scanning billions of random peptides, we accurately map binding specificity for approximately half of the over 330 PDZ domains in the human and Caenorhabditis elegans proteomes. The domains recognize features of the last seven ligand posit...
متن کاملStructures of the human Pals1 PDZ domain with and without ligand suggest gated access of Crb to the PDZ peptide-binding groove
Many components of epithelial polarity protein complexes possess PDZ domains that are required for protein interaction and recruitment to the apical plasma membrane. Apical localization of the Crumbs (Crb) transmembrane protein requires a PDZ-mediated interaction with Pals1 (protein-associated with Lin7, Stardust, MPP5), a member of the p55 family of membrane-associated guanylate kinases (MAGUK...
متن کاملNHERF1/EBP50 head-to-tail intramolecular interaction masks association with PDZ domain ligands.
Loss of cell polarity is one of the initial alterations in the development of human epithelial cancers. Na(+)/H(+) exchanger regulatory factor (NHERF) homologous adaptors 1 and 2 are membrane-associated proteins composed of two amino (N)-terminal PDZ domains and an ezrin-radixin-moesin (ERM)-binding (EB) carboxyl (C)-terminal region. We describe here an intramolecular conformation of NHERF1/EBP...
متن کاملComposition and function of PDZ protein complexes during cell polarization.
Complexes consisting of PDZ proteins have been implicated in a variety of cellular processes. In recent years, it has become increasingly clear that PDZ proteins play essential roles during the establishment of spatial asymmetry in various metazoan cell types such as epithelial cells. Epithelial cells possess asymmetry with respect to the apicobasal axis reflected by the differential distributi...
متن کاملOn the Role of PDZ Domain-Encoding Genes in Drosophila Border Cell Migration
Cells often move as collective groups during normal embryonic development and wound healing, although the mechanisms governing this type of migration are poorly understood. The Drosophila melanogaster border cells migrate as a cluster during late oogenesis and serve as a powerful in vivo genetic model for collective cell migration. To discover new genes that participate in border cell migration...
